Mark Tune was born before his father's will was proven in 1677. He was the son of James Tune and Anne and the youngest of six siblings. In his father's will Mark received along with his brother Caster "My other seat of land which John Massy now liveth on to be equally divided between them."

Mark married Elizabeth Lewis about 1698. She was the daughter of John Lewis and Mary Garner. They had at least seven children.

Mark wrote his will on September 21st, 1717 and it was probated in Richmond County court on June 4th, 1718. Elizabeth must have died before the will was written because there is no mention of a wife. Mark divided his land among his four oldest sons and left beds and furniture to his youngest son and two daughters.
