Elizabeth Brooke was born about 1503 in Cobham, Kent, England. She was the daughter of Sir Thomas Brooke and Dorothy Heydon. Thomas was the 8th Baron of Cobham, he fought in the war against France in 1513. He was knighted in 1514 and served in the King's parliament from 1514 to 1523.

Elizabeth married Thomas Wyatt about 1520 or 1521. They had only one child. Thomas was believed to have had several affairs that damaged their relationship, and they informally separated around 1525. His most famous rumored affair was with Anne Boleyn. In 1536 when Anne was married to King Henry VIII, Thomas was arrested and charged with treason. He was held in the Tower of London along with Anne and probably witnessed her execution. Thomas was eventually pardoned. Thomas also accused Elizabeth of being unfaithful and having an affair with King Henry. Thomas took a mistress before 1537, Elizabeth Darrell, and had three more sons. At one point, Elizabeth Brooke's brother, George, contacted Thomas Cromwell, a friend of Thomas Wyatt, and asked him to make sure Thomas provided for his wife.

Thomas again fell from King Henry's favor in 1541 and was again charged with treason, this time for verbal treason by wishing for the death of the king. It is said that Thomas confessed his guilt and received a pardon on the condition that he return to his wife, Elizabeth. Thomas died while traveling in Dorset the following year.

Elizabeth remarried to Sir Edward Warner. They had three children but none survived infancy. Elizabeth died about 1560.
