Thomas was the son of Thomas Wyatt Sr and Elizabeth Brooke. Thomas Sr was a poet and an ambassador in the court of King Henry VIII. He was rumored to have had an affair with Anne Boleyn which led to her execution and him being imprisoned in the Tower or London and charged with treason. Jane and Thomas went on to have ten children. After the death of Thomas Sr, Thomas Jr contributed to a book of his father's poetry by transcribing letters of advice from his father including advice to study moral philosophy and live contentedly with his wife, following the good example of his grandfather rather than the past behavior of his father. The book still survives today and is held by the British Library.
Thomas inherited many lands from his father but had to sell several estates because of the debts he also inherited. Thomas became a soldier and quickly rose through the ranks. He was knighted in 1547 and served as Sheriff of Kent in 1550.
In January of 1554 Queen Mary I announced her intention to Mary King Philip of Spain. Thomas and several of his friends took offense to that decision and met to plan a resistance but several were arrested and Thomas became the leader of the movement that came to be called Wyatt's Rebellion. Soon after Thomas left home for the last time with a force of 4,000 men to march on London. The rebellion was put down and Thomas was captured and charged with treason. He was executed on April 11th, 1554.
Because Thomas was charged with treason his lands and title were taken by the crown. Queen Mary paid Jane, an annuity of 200 marks as an act of compassion and returned a small portion of her dower lands. When Queen Elizabeth I came to power in 1558 she reversed the remaining penalties against the family.
Jane is not known to have remarried and thought to have lived until about 1600 and died at Allington Castle in Boxley.
